Mapping the exposome of mental health: exposome-wide association study of mental health outcomes among UK Biobank participants. Dissecting the exposome linked to mental health outcomes can help identify potentially modifiable targets to improve mental well-being. However, the multiplicity of exposures and the complexity of mental health phenotypes pose a challenge that requires data-driven approaches. Guided by our previous systematic approach, we conducted hypothesis-free exposome-wide analyses to identify factors associated with 7 psychiatric diagnostic domains and 19 symptom dimensions in 157,298 participants from the UK Biobank Mental Health Survey. After quality control, 294 environmental, lifestyle, behavioral, and economic variables were included. An Exposome-Wide Association Study

Putting the exposome into practice: An analysis of the promises, methods and outcomes of the European human exposome network. Contemporary research on the exposome, i.e. the sum of all the exposures an individual encounters throughout life and that may influence human health, bears the promise of an integrative and policy-relevant research on the effect of environment on health. Critical analyses of the first generation of exposome projects have voiced concerns over their actual breadth of inclusion of environmental factors and a related risk of molecularization of public health issues. The emergence of the European Human Exposome Network (EHEN) provides an opportunity to better situate the ambitions and priorities of the exposome approach on the basis of new and ongoing research. Network-Based Identification of Key Toxic Compounds in Airborne Chemical Exposome. Air pollution is a leading contributor to the global disease burden. However, the complex nature of the chemicals to which humans are exposed through inhalation has obscured the identification of the key compounds responsible for diseases. Here, we develop a network topology-based framework to identify key toxic compounds in the airborne chemical exposome. Using cardiovascular diseases (CVDs) as a model disease, we found that toxic network modules of various compounds are closely linked to the modules of CVDs. The proximity of compound target modules to disease modules can indicate the extent of toxicity induced by the compounds. By integrating mass spectrometry-based external exposure concentrations and machine

Impact of the Adverse Social Exposome on Survival in Individuals With Amyotrophic Lateral Sclerosis. An adverse social exposome negatively affects many diseases, but its association with amyotrophic lateral sclerosis (ALS) survival is unknown. This study examined the association between the social exposome measure Area Deprivation Index (ADI) and ALS survival. This is a retrospective analysis social exposome, as measured by ADI, associates with poorer ALS survival. Because this is a single-center study, replication in other cohorts is encouraged. Further research is needed to understand the underlying mechanisms, which could influence ALS clinical care.

An Enhanced Protocol to Expand Human Exposome and Machine Learning-Based Prediction for Methodology Application. The human exposome remains limited due to the challenging analytical strategies used to reveal low-level endocrine-disrupting chemicals (EDCs) and their metabolites in serum and urine. This limits the integrity of the EDC exposure assessment and hinders understanding of their cumulative health effects. In this study, we propose an enhanced protocol based on multi-solid-phase extraction (multi-SPE) to expand human exposome with polar EDCs and metabolites and train a machine learning (ML) model for methodology prediction based on molecular descriptors. The protocol enhanced the measurement of 70 (25%) and 34 (12%) out of 295 well-acknowledged EDCs in serum and urine compared

De Novo exposomic geospatial assembly of chronic disease regions with machine learning & network analysis. Determining spatial relationships between diseases and the exposome is limited by available methodologies. aPEER (algorithm for Projection of Exposome and Epidemiological Relationships) uses machine learning (ML) and network analysis to find spatial relationships between diseases and the exposome in the United States. Using aPEER we examined the relationship between 12 chronic diseases and 186 pollutants. PCA, K-means clustering, and map projection produced clusters of counties derived from pollutants, and the Jaccard correlation between these clusters with chronic disease geography (defined as groups of counties with high chronic disease prevalence rates) was calculated. Inhaled pollutants of the Gero-Exposome and later life health. Inhaled air pollutants (AirP) comprise extraordinarily diverse particles, volatiles, and gases from traffic, wildfire, cigarette smoke, dust, and various other sources. These pollutants contain numerous toxic components which collectively differ in relative levels of components, but broadly share chemical classes. Exposure and health outcomes from AirP are complex, depending on pollutant source, duration of exposure, and socioeconomic status. We discuss examples in the current literature on organ responses to AirP, with a focus on lung, arteries, and brain. Some transcriptional responses are shared. It is well accepted that AirP contributes to Alzheimer's disease and other neurodegenerative conditions in the Gero-Exposome. However

Adopting Mechanistic Molecular Biology Approaches in Exposome Research for Causal Understanding. Through investigating the combined impact of the environmental exposures experienced by an individual throughout their lifetime, exposome research provides opportunities to understand and mitigate negative health outcomes. While current exposome research is driven by epidemiological studies that identify associations between exposures and effects, new frameworks integrating more substantial population-level metadata, including electronic health and administrative records, will shed further light on characterizing environmental exposure risks. Molecular biology offers methods and concepts to study the biological and health impacts of exposomes in experimental and computational systems

Geospatial Science for the Environmental Epidemiology of Cancer in the Exposome Era. Geospatial science is the science of location or place that harnesses geospatial tools, such as geographic information systems (GIS), to understand the features of the environment according to their locations. Geospatial science has been transformative for cancer epidemiologic studies through enabling large-scale environmental exposure assessments. As the research paradigm for the exposome, or the totality of environmental exposures across the life course, continues to evolve, geospatial science will serve a critical role in determining optimal practices for how to measure the environment as part of the external exposome. The objectives of this article are to provide a summary of key concepts, present a conceptual

Exposome and basal cell carcinoma: a multicenter case-control study. While ultraviolet radiation (UVR) present in sunlight is recognized as the main etiological agent of skin cancer, the most frequent form of which is basal cell carcinoma (BCC), other exposome factors like pollution, diet, and lifestyle may also contribute. This study aimed to investigate the association of BCC and exposome (P = 0.03). Higher linolenic acid intake and lower coffee consumption were the only dietary variables associated with BCC (P < 0.05). Statistical significance for all the aforementioned variables was maintained in the multivariate analysis (P < 0.05). The study found a significant association between BCC and multiple exposome-related factors in addition to chronic sun exposure in the Spanish

Hair-Derived Exposome Exploration of Cardiometabolic Health: Piloting a Bayesian Multitrait Variable Selection Approach. Cardiometabolic health is complex and characterized by an ensemble of correlated and/or co-occurring conditions including obesity, dyslipidemia, hypertension, and diabetes mellitus. It is affected by social, lifestyle, and environmental factors, which in-turn exhibit complex . We identified six exposures that were jointly explanatory of cardiometabolic health as modeled by six complementary traits, of which, we identified strong associations between hexachlorobenzene and trifluralin exposure and adverse cardiometabolic health, including traits of obesity, dyslipidemia, and hypertension. This supports the use of this type of approach for the joint modeling, in an exposome
