Upregulations of Clcn3 and P-Gp Provoked by Lens Osmotic Expansion in Rat GalactosemicCataract Lens osmotic expansion, provoked by overactivated aldose reductase (AR), is the most essential event of sugar cataract. Chloride channel 3 (Clcn3) is a volume-sensitive channel, mainly participating in the regulation of cell fundamental volume, and P-glycoprotein (P-gp) acts as its modulator. We aim to study whether P-gp and Clcn3 are involved in lens osmotic expansion of galactosemiccataract. In vitro, lens epithelial cells (LECs) were primarily cultured in gradient galactose medium (10-60 mM), more and more vacuoles appeared in LEC cytoplasm, and mRNA and protein levels of AR, P-gp, and Clcn3 were synchronously upregulated along with the increase of galactose concentration. In vivo, we focused
Diosgenin, a Novel Aldose Reductase Inhibitor, Attenuates the GalactosemicCataract in Rats To seek efficient aldose reductase inhibitors (ARIs) with excellent in vitro and in vivo biological activities against rat galactosemiccataract. The method was firstly optimized to screen strong ARIs from nonoriented synthetic compounds and natural extracts. Then, diosgenin was assessed on osmotic . In vivo, diosgenin delayed lens opacification, inhibited the increase of ratio of lens weight to body weight, and decreased AR activity, galactitol level, and AR mRNA expression, especially in the diosgenin drinking (0.1%) group. Diosgenin was an efficient ARI, which not only significantly decreased the LECs' osmotic expansion in vitro but also markedly delayed progression of rat galactosemiccataract
Cytoplasmic phase separation in formation of galactosemiccataract in lenses of young rats We have determined the age dependence of the characteristics of the cytoplasmic phase separation of lenses from normal and galactosemic young rats. In the normal lens, the temperature at which the phase separation occurs decreases monotonically with age. In the lenses of rats fed with a high galactose diet
